SPIN® technology for better IVD antibodies

Antibodies are a vital reagent for sensitive immunoassays. SPIN®, proprietary platform uses microfluidic technology to recognize single plasma cells and isolate only the antigen specific cells. By combining the power of rabbit’s unique immune system and SPIN® technology’s efficient isolation of antigen specific plasma cells generates large number of ELISA positive high affinity antibodies. The large number of ELISA positive clones can be screened for desired binding specificity and kinetics suited for intended immunoassays.

Screening Data

Highly specific antibodies from plasma cells are generated against nucleocapsid and spike protein of SARS-CoV-2 using SPIN® technology. The recombinant antibodies developed have no cross reactivity against human coronavirus nucleocapsid protein.

Neutralization Assay

The two clones S185 and S422 display the best neutralization against the SARS-CoV-2 pseudo virus.The neutralization IC50 values are recorded in micro­ grams per millimeter.

Neutralization assay Anti- RBD clones S185 and S422 were assessed for neutralization activity on VSV pseudotyped virus carrying a spike protein of SARS,MERS, SARS-CoV and SARS-COV-2 mutant (D614G) respectively. D614G mutant is the prevalent type in US now. The results indicate that these antibodies have good neutralization activity to wild type and D614G mutant of SARS-COV-2 pseudoviruses, but no cross­ neutralization activity to SARS and MERS pseudoviruses.
