Background information
Carboxyfluorescein (FAM) has become a go-to hapten for oligo-based diagnostics, largely because anti-FAM antibodies bind it with high specificity, giving assays a clean, low-background signal. Its use is especially common in quantitative PCR, where FAM-labeled primers or probes allow real-time monitoring of amplification. That same labeling chemistry carries over well into lateral flow formats: after amplification, FAM-tagged amplicons are captured at the test line by anti-FAM antibody, producing a simple visual result that needs no lab equipment to read. Because FAM shares its core structure with FITC, cross-reactivity between anti-FAM and anti-FITC reagents is common and should be checked for a given kit rather than assumed away. Paired with a second hapten such as biotin in a sandwich format, FAM-based lateral flow assays deliver fast, low-cost molecular detection suited to research, clinical, and point-of-care environments alike.
